For the first question, I think the idea is that only the first try counts (you can "practice" afterwards, but the score won't count).
I observe the stairs bit definitely worked. As far as Challenge Of The Week goes, I think it's an excellent concept. Though I did die pretty quickly. score 361, lvl 4

Is the scoring system working properly for the challenge?
I attempted it just now and died to a tension room of green hags on I:9 after reaching char lvl 5.
I got negative score, around -182. Is this normal?
Green hags curse you/your equipment, which may input some negative variables in the score, iirc
Challenge game is working fine for me as regards individual scores. First game of the week is scored, and all others are practice runs for the next one.
Highest score I've got so far is just over 1000.
I just had a PC die on level 3 and I:3 to a mixed tension room straight off the stairwell that included werewolves among other things - is this overlevelled as well?
